An ethical leader is a leader who has good virtue characteristics as a human being that displays good quality leadership that is thoughtful of others that shows caring and kindness and believes in fairness to others. A good ethical leader encourages others to do the right thing and lead others in the right directions of making the right decisions. This paper contains virtues and vice on Bill Gates charity organization and business affairs. The evidence is shown that Bill Gates is a man of many virtues proven by a charity organization called Bill and Melinda Foundation. However, Gates has proven to be a strong ethical leader there are some who believe otherwise when it comes to unethical practice as a businessman of some of the top largest company. The goal of this paper is to focus on effective leadership of Bill Gates should be handled in an ethical manner on managing business and employees with consideration and respect. Virtues of Bill GatesThere are two virtues about Bill Gates that best describes as an effective ethical leader of Bill and Melinda Foundation. The first virtue is sincerity is one of the characteristic traits of Gates. According to Oxford Dictionary, the definition of sincerity is the quality of being free from pretense, deceit, or hypocrisy (oxforddictionary.com). Sincerity meant as being generous and kind and caring to others such as individuals who in need of help in all lives. The Bill and Melinda foundation is a charity foundation that helps and improves poverty, education, children well-being, and health. Gates donated billions of dollars towards foundation charity that contributes to people around the world. The foundation has helped proven the sincerity virtue of Gates moral characteristic traits in the most critical issues that needed the most. There are many evidences that Gates invested into healthcare of 4.2billion dollars and 1.5billion for the homeless Lancet, (2007). Gates has well proven to help save and change world by creating Bill and Melinda foundation which is the world's largest charity foundation.The second virtue is wisdom that Gates has proven to be good moral character by building a successful foundation of charity by organizing, gathering information, and demonstrating knowledge of what are critical issues that needed to be addressed to make the world a better place. The Oxford dictionary gives the definition of wisdom as the quality of having experience, knowledge, and good judgment; the quality of being wise (oxforddictionary.com). Wisdom is meant as having the knowledge and good decision making for an effective benefit. There are two types of wisdom such as theoretical and practical. The theoretical wisdom is explained as to understanding and comprehending. According to Brusseau (2012), describes practical or sometimes called prudence as the learned knowledge that being able to apply it in real life. Bill Gates of Vice as InsensibilitySome believe that Gates has another side that made Gates one of the richest entrepreneurs in the world. Bill Gates has a number of successful businesses. One of the companies, in particular, is a waste management company called Republic Service. Republic Service that Gates own is one of the world's second-largest companies in America. Accusations have been made against Gates has mistreated the employees of Republic Service with unethical practice and standards. The three major concerns that workers are disputing over retirement funds being taken, unpaid overtime, and illegal abandoning contracts employee already agreed that was with the union. This caused employees to protest and being lockout Republic Service.The term insensibility is one of example that applies to Gates on this matter. Assuming that Gates is guilty of this unethical behavior, this is an act of no consideration for the employees if employees' benefits were taken that employee had worked for many years at the waste management company. The dictionary gives the definition of insensibility as incapable of feeling or perceiving; deprived of sensation; unconscious, as a person after a violent blow (dictionary.com). Assuming that allegations are true this unkindly act of unethical practice, then evidence has shown that Gates should show more of effective ethical practice and standards towards employees with consideration and respect. Conclusion Bill Gates is believed to be a good example of an ethical leader. Bill Gates is a self-made billionaire who was the creator of Microsoft and other software. Gates had changed the lives of people all over the world with the innovation of Microsoft. Bill Gates has a very successful resume and continues to pursuing on helping save the world which is an inspiration to everyone. Bill Gates has a way of touching people hearts because of the concerns of many critical issues that the world is faced with. Gates has a way of tackling major issues that no one wants to deal with let alone donate an unlimited amount of funding so give thanks to Gates.Despite what others are saying, many have an issue with Gates the billionaire money which shouldn't matter. After all, there are always some allegations or accusations that are being made about someone especially like Bill Gates that is rich and famous can't expect to have enemies. Overall, what should matter if Gates behaving unethical manner. The value of employees should be held to the highest standards of ethical behavior with the utmost respect? Let's concentrate on what all said to be practiced and what all is said to be preached. Its similar to the purchase invoice and includes things like VAT and discounts. Also it includes the description of the product. The sales invoice is different because its’s only issued when a good has been delivered and has not paid for unless the customer has paid immediately. Delivery Note (see Example 7) A delivery note is given to the customer when the goods are delivered. The documents tells you what has been delivered and how many has been delivered. Also on it is the customers address so its similar to the goods received note. This document is a security measure so the right goods end up in the right place. The delivery note is given to the driver of the truck who deliver the goods and is then passed on to the customer. Sales Credit Note (see Example 8) This document is used by the customer in the event of an error in the supplied goods. For example if you bought a pair of shoes returned home and found a tear in them you can ask for a refund or accept a sales Credit Note. This would allow you to return to the shop in the future and exchange the note for a good or goods of the same value. The credit note is issued by the supplier and is sent to the customer. Statement of Account ( see Example 9) A statement of account is sent by a supplier to a customer. It states all transactions made in the month by the customer, any money outstanding, the total amount of money due on each invoice and the total balance which has to be paid. Remittance Advice (see Example 10) A remittance advice slip will often accompany the statement of account form. It is sent with cheque by the customer. This slip helps the supplier accurately process the payment when it is received. The supplier issues the remittance advice slip and the customer makes use of it by matching it with the invoice or statement of account and then sends it back with the cheque or other payment, this makes it easier for the supplier to match up cheque to invoices When you pay for something in cash you need some sort of proof you’re bought it. The proof you need is to collect a receipt. This is needed if you want to make a refund or if you want to exchange your product for something else. The receipt is written proof of your purchase and should include the following:- The date, Receipt number, Name of company, Description of the product, Amount paid (inc. VAT). Some of the oncology patients were severely neutropenic. This poses a greater risk for infections since the white blood cells needed to fight infection is very low. It has been noted that patients with infectious diseases such as tuberculosis are being nursed on the same ward, along with burn patients and HIV patients. This issue is significant to oncology nursing and cancer care since it addresses the practices of nursing and also plays an important part in patient outcomes. It has been estimated that each year in the United States there were sixty thousand cases of cancer patients hospitalized with neutropenia. About two thirds of that number has reported infections and the estimated inpatient mortality rate of those patients is seven percent (Segal, 2008). Infections also cost hospitals a lot of money that could be spent on other health issues. It was estimated that infections have added thirty billion dollars to the US hospital cost while consumers also have to pay part of the cost (McCaughey, 2005). A search was carried out using internet data bases and articles were found which lends support to the use of barrier nursing and other methods as effective tools in infection control. Barrier Nursing is nursing a patient so as to erect a barrier to the passage of infectious pathogenic organisms between the contagious patient and other patients and staff in the hospital, and thence to the outside world (Last, 2010).It involves the use of having the patient isolated in a separate room and if not possible they are screened on an open ward. The nurse would wear a gown, mask and gloves when attending to the patient and observe strict hand washing and aseptic techniques (Last, 2010). Reverse barrier nursing or protective isolation is where the susceptible patient is isolated to prevent them from getting infections. It involves limiting the number of staff and visitors that interact with the patient especially if they are ill. It also involves limiting attending to a contagious patient then attending to the immunocompromised patient. All staff should be aware of the precautions and the doors should be appropriately labeled (The Leeds Teaching Hospital, 2003). According to the American Cancer Society (2009), neutropenia is described as a deficiency of neutrophils. Neutrophils are the largest number of white bloods cells in the body and are crucial to the bodys defense system. Some cancers such as the lymphomas and some leukemias directly affect the immune system and causes low blood counts. In other cases the cancer treatment received such as chemotherapy and radiation results in damage to the immune system (American Cancer Society, 2009). When the immune system is damaged or suppressed it is referred to as immunosupression. When this occurs the white blood cells especially neutrophils are unable to fight off infections and the person is highly susceptible to getting an infection. Infections can be caused by bacteria, viruses, parasites or fungi (American Cancer Society, 2009). Some studies have suggested that proper hand hygiene by the patient and the nurse along with immunization, antifungal and antibacterial prophylaxis have show to be the most effective methods of infection control. Avoiding contact with persons who are ill with respiratory illnesses has also been suggested (Zitella, Friese, Hauser, Gobel, Woolery, OLeary and Andrews, 2006). The Center for Disease Control recommends hand washing as the most important means of spreading infection especially in susceptible populations such as the immunocompromised. It is also important between different patients in high risk units such as between cancer patients and HIV patients (LeTexier, 2000). Other studies have suggested that barrier nursing is needed for all immunocompromised patients. Some infections are seen mostly in certain disease populations for example Candida infections are found in many HIV patients. However the question of whether neutropenic patients should be housed close to HIV patients needs to be further researched. Rather strict hygiene practices along with prophylactic treatment have been suggested (Lortholary Dupont, 1997). It is suggested that all clinical staff and other health care workers are periodically trained in infection control and there should be consistent evaluation of techniques being used. The nurse is in a unique position to ensure that infection spread is reduced by carrying out and monitoring aseptic practices; ensuring the environment is clean to prevent spread of micro organisms and to protect the patient from ill staff and visitors (Collins, n.d). Some studies have also indicated that there is little evidence that reverse barrier nursing is effective especially where there is no laminar air flow or HEPA-filtered rooms available. However some studies indicate that the filtered rooms can reduce morbidity and mortality in patients with leukemia (Seshadri Baumann, 2008). The use of reverse isolation of any type for patients with solid tumors who are neutropenic is discouraged since there are no studies to prove it is helpful rather it can cause anxiety and confusion for patients, their families, and healthcare workers (Seshadri Baumann, 2008). After reviewing the literature the question can be answered that neutropenic patients can be effectively nursed on these wards with the collaboration of all the members or the health care team in carrying out proper hygienic methods.

Freidman summarizes the three globalizations that have occurred in the world. The first one in 1492-1800, making the world to a medium size. Second one in 1800 to 2000 making the world to a size small and third one starting in 2000 making the world flat after all these years of developing technology causing to become an integrate and flat world. Freidman’s 10 events and forces starting in Nov. 9, 1989 with Berlin Wall’s falling down mention how the world get flattened so fast. He mentions in these 10 events the Netscape moment offering the dot-com and connecting people in a way never before. This enable Indian engineers which years back when the world wasn’t globalized didn’t had the opportunity to compete in a position with an American to be enlisted for outsourcing. With these events the world suddenly began outsourcing, offshoring, open-sourcing, insourcing, supply-chaining and informing people and connecting them, showing the world a whole new idea: that we are living in a globalize world. The final event he called it â€Å"the steroids† which is the wireless access that enables you to connect with every person you want from anywhere with any device. Which is contrary to what Ghemawat says, â€Å"People are getting more connected but they aren’t connecting† (pg. 58 Ghemawat, 2005). The fact is that people are spending time chatting with family and friends that meeting someone to start a flat world. It is true that not everyone has the access to technology, but is a fact that each day more people get connected and collaborating to this than ever before in history’s life, this is the first step for a globalize world. Americans had become lazier in innovating probably because they never thought that globalization would be knocking in their front doors. â€Å"This is not a test† Freidman says, is already happening and the soonest you prepare for this the soonest you will be part of this flattened world. â€Å"Why the World Isn’t Flat† by Pankaj Ghemawat is just the opposite as everything Freidman tells, making an exaggeration to all this presumption of the world being flat. In the article he states facts proving we are far away from a fully globalized world. He evidences this by stating, â€Å"90% of all phone calls, web traffic and investment is global. † Freidman’s 10 forces seem persuasive but Ghemawat’s The 10 Percent Presumption proved him wrong. In these presumptions it shows that immigration, phone calls, direct investment, and others don’t even exceed the 10% in levels of internationalization across industries. Patents and portfolio investment barely exceeds the 10% and trade hardly beats the 20%. Even with trades being the highest you cannot have a globalize world with that percentage shown. For Ghemawat all of this flat world that Freidman says contains an exaggeration maybe in order to sell books and that we in fact are more cautious and prefer to be in a protectionism world. Although major revolutions as the Internet has improved the way we globally integrate, the barrier where globalization’s principles: language, geography and distance doesn’t matter is still not functioning at all. Russia’s local service Yandex beating up Google because they don’t have the ability to understand the complexities of the Russian language proves that we are not living in a globalize world. He also states that there is a fear of becoming a globalize world as tendency is supporting more protectionism and â€Å"that globalization may be incompatible† While Freidman’s posture is mentioning that a flat world involves everyone around, as a video-conference with the entire supply chain of the company and different employees all at once. You need to leave protectionism and a fear of being globalized and knowing you’re in a flat world when your office has eight different clocks. Ghemawat mentions that people like Friedman are describing globalization, as a world that doesn’t exists. The truth is for Freidman that people don’t know but we are actually living a globalization that is shown by stating a fact that China graduates twice students with bachelor’s degree than US. â€Å"America is falling behind† he says, but the truth is that everyone is falling behind if they don’t realize that the world is becoming more flat and less round. â€Å"The World is Spiky† by Richard Florida mentions that the world is more harrowing and less flat as Freidman says. He refers spiky with a map as the different peaks around the world and how they aren’t constant but significantly diverse that you cannot call it a flat world as Freidman. There had been a growth in countries with the major cities but they are leaving their other cities behind this process of globalization. Florida says this spiky globalization in big countries like China are concentrated their production, innovation in their rural regions leaving the urban outside of this making them have conflicts. Instead of a flatter world is becoming a spiky world with peaks dispersed all around it but in an inconsistent way. Instead of raising spikes, there needs to be valleys to have equality in their peaks for a flat world to come and he’s arguing why Freidman left this gap between the poor and rich behind. Also the article by Matt Taibbi â€Å"Flathead† for the New York Press, says he is not surprised about what Freidman says because he always continues to fail his metaphors in this case the world being flat. Arguing in favor of the exaggerations Ghemawat said about Freidman and the harrowing the world is as Florida. I believe that Freidman exaggerated of the world being flat but also I believe this is just what we need in order to have a motivation and not getting stuck in the laziness and comfort. Maybe this fear of globalization people have, is because they know that they’re living in a globalize world where they can be left behind if they don’t realize the fact that today the world is becoming more flat and less round or spiky.

It has been seen that a lot of people migrate to new cities or new countries these days. They may face a number of changes in their new life, including climatic change, changes in language and the way of eating food. The history of African American music has been characterized by a mixture among various forms of music. Country blues, urban blues, New Orleans Jazz, Bebop, big-band jazz, and rhythm and blues, have all influenced each other profoundly. These influences flowed back and forth among the various forms. But, black gospel music had only a very limited effect on popular styles, until a few church-trained artists, such as Sam Cooke and Ray Charles, began to incorporate gospel styling into their popular work. The result is usually described as â€Å"soul music,† a mix of blues, rhythm and blues, and gospel voices. But, if Ray Charles was one of the â€Å"originators† of soul music, â€Å"Aretha Franklin reshaped it,† by bringing even more of her gospel background to bear on secular love songs (Wade and Picardie 27). By combining popular elements with her stunning voice, her great musicianship, and the feeling for a song that she learned in church, Aretha became one of the greatest soul singers to ever live. Aretha Franklin is a well-known pop, R&B, and gospel singer. She has been nicknamed â€Å"The Queen of Soul† and is an internationally known artist and a symbol of pride in the African American community. Her popularity soared in 1967 when she released an album containing songs â€Å"I Never Loved a Man†, â€Å"Respect†, and â€Å"Baby I Love You. † Throughout her career she has achieved fifteen Grammy Awards, Lifetime Achievement Award, National Academy of Recording Arts and Sciences Legend Awards, and many Grammy Hall of Fame Awards. In 1987 she became the first woman inducted into the Rock & Roll Hall of Fame. Time magazine chose her as one of the most influential artists and entertainers of the 20th century. She sang at Dr. Martin Luther King’s funeral and at former President Bill Clinton’s inaugural party. Although she has all these accomplishments and awards there are other reasons that have driven Franklin to fame and landed her on the front cover of Time magazine on June 28, 1968. The reasons I believe allowed Aretha Franklin to become so successful are the following: Her family’s involvement with religion, the inspiring people that surrounded her, and the pain she suffered. It is clear that because her family’s involvement with religion would be one reason why Aretha Franklin became as famous as a Gospel singer. Some people would say that her love for religion is unbelievable, but after researching her childhood it is very believable. Her father, Reverend Clarence LaVaugh Franklin lived in Shelby Mississippi and preached while living the life of a sharecropper. As soon as he had enough money, he would move to Memphis, Tennessee to become a pastor of two churches. After a couple of years he attended LeMoyne College, and he studied Education and English Literature. With his education he was able to bring a more liberal view to his preaching’s. Then he moved the family to Buffalo, New York. When he had the resources, he moved the family again to Detroit, Michigan were he settled and became a pastor of a churched called New Bethel Baptist Church. He quickly became one of the most famous pastors in the city of Detroit. Aretha was two years old when they made their final move, she would grow up here and grab the emotion of Church and incorporate it into her music. Aretha Franklin’s mom, Barbara V. Skaggers, served as choir director and pianist. Aretha describes her mom as â€Å"a Superb singer, her voice was clear and distinctive†. (Franklin and Ritz, 6) Her parents taught her how to sing with great pride. This was a big issue because the late ‘50s, early ‘60s was a time of turmoil for African Americans. Her father especially tried to instill pride into her. He was a Civil Rights activist and he was a close colleague with Dr. Martin Luther King. With her parents keeping her involved in Church she was bound to become one of the world’s greatest singers. At around age 12, the father recognized Aretha’s talent as a singer. So he took her on the road with his traveling gospel show. This was important because it shows the kind of support Aretha received from her family. It was said, â€Å"She was a spellbinding performer at the age of fourteen. † (Franklin, 3) So her family really supported and inspired her to become a gospel singer. What also made her a great artist was that she had inspiring people all around her. Aretha grew up in Detroit which at the time was a rousing city or a city of hope for the African Americans running away from the brutality of the South. Though Detroit still had its problems such as race riots, many famous musicians grew up there. Also since New Bethel Baptist Church was so prominent, many musicians and political leaders used Reverend Franklin’s pulpit as a platform to sing or speak to the Black’s of Detroit. Aretha was introduced to classical music by Smokey Robinson’s sister Sylvia Burston. She listened to well known local DJ’s like Ruth Brown and Senator Brystal Brown. When Aretha was younger, she would ride her bike to the local park, and on her way home she would stop by a night club where you could here B. B. King perform. She says, â€Å"You could hear the soft sound of his guitar all the way to the sidewalk (Franklin and Ritz, 22). National and local political leaders would give there speeches. Speakers such as Dr. Adam Clayton Powell, Sr. , Dr. Martin Luther King, and Reverend Jesse Jackson would speak powerfully to the church. Aretha was directly influenced by Miriam Anderson, Sammy Davis, and Roy Wilkins. Detroit was overflowing with talent and speakers which I believe also contributed to Aretha’s success. Pain was probably what really drove Aretha Franklin’s success. As stated before, Franklin’s family was highly religious and was continually involved in the Church. But that doesn’t mean that she hadn’t been through a tremendous amount of pain. Early in life her mother and father got a divorce. The father was better suitable to raise Aretha and her four Siblings. The mother moved to Buffalo, New York and tried to make regular visits to see her children. Brown V. Louisiana Essay During the 1960s, many African-Americans believed that civil rights should become a national priority. Young civil rights activists brought their cause to the national stage and demanded the federal government assist them and help resolve the issues that plagued them. Many of them challenged segregation in the South by protesting at stores and schools that practiced segregation. Despite the efforts of these groups and Supreme Court rulings that ordered the desegregation of buses and bus stations, violence and prejudice against African-Americans in the South continued Meyer, F. S. , 1968). In the 1960s many things were off limits to African-Americans. They werent revered as equals and suffered greatly because of it. Theres an unfamiliar case to most that took place in Louisiana that helped shaped the use of public facilities for all people. This case is known as Brown v. Louisiana. The Audubon Regional Library in Clinton, Louisiana, Parish of East Feliciana did not serve blacks. Blacks, at that time, were expected to use one of two bookmobiles. The red bookmobile served whites and the blue bookmobile served blacks. On March 7, 1964, ive young African-American males entered the adult reading room and one of the men, Brown, requested a book called, The Story of the Negro, by Arna Bontemps. The assistant librarian checked the card catalogue and discovered that the library did not have the book. She told Brown that she would request it from the state library and he could either have it mailed to his home address or he could pick it up from the bookmobile. After the men had been given the news about the book they sat down quietly. After the men failed to leave the library, the assistant librarian requested that they go. They did not. Brown sat down while the others stood nearby. The assistant librarian then went to the head librarian who requested them to leave as well. Again, they did not. A few moments later, the sheriff arrived and requested that they leave again, and again, they did not. The sheriff arrested them and charged them with the intention to provoke a breach of peace and failure to leave a public building when ordered to do so (Coates, R. , 2005). The five men were tried and found guilty. Brown was sentenced to pay $150 for court costs or spend 90 days in Jail. The four other men were sentenced to $35 for court costs or 15 days in ail. Under Louisiana law, the convictions werent appealable therefore; their requests for discretionary reviews were denied. The Supreme Court granted certiorari. A certiorari is an extraordinary privilege injunction granted in cases that otherwise would not be entitled to review. In writing for the majority, Justice Fortas first examined whether the protesters could be convicted for refusing to leave the library. He concluded that they could not since their protest was peaceful and blacks could not be denied access since whites were allowed inside as well. He reviewed the onduct of the men and felt that this had no merit either. The state argued that the men were proving their intent to disturb the peace and upset the librarian. Justice Fortas concluded that the arrest was a violation of the mens First and Fourteenth Amendment rights that guarantee freedom of speech and assembly and the right to opposed this opinion and took to issue with the majoritys reasoning. He disagreed that the Constitution prohibits any state from making sit-ins or stand-ups in public libraries illegal. Second, Black argued that the previous breach of the peace cases in Louisiana differed from Brown v. Louisiana. Previously there had been several other situations where there were peaceful demonstrations over discriminatory practices. Garner v. Louisiana (1961) involved a sit-in at a lunch counter to protest service for whites only. In Taylor v. Louisiana (1962) blacks again protested the presence of bus depot that was for white customers only. In Coxv. Louisiana (1965) a man led a demonstration near the courthouse and Jail to protest the arrest of other demonstrations. Each of the protests, along with Brown v. Louisiana, was all orderly and peaceful and was over discriminatory practices that denied the protesters rights hat were guaranteed to them under the Constitution. Justice Black opposition was joined by three other Justices. They argued that the First Amendment did not guarantee to any person the right to use someone elses property even that owned by the government and dedicated to other purposes. On Wednesday, February 23, 1966 the decision was made; 5 votes for Brown and 4 against him (Coates, R. , 2005). The young men won! The Courts ruling in this case, along with the others, proved vital to the Civil Rights struggles and also to the Vietnam War protests that would follow. Indeed, without these rulings the 1960s and early 1970s may have been a completely different period in time, especially when it comes to the Civil Rights movement. Uses of DNA Technology Introduction DNA typing was first used in Great Britain for law enforcement purposes in the mid- 1980s and has revolutionized forensic science and the ability of law enforcement to match perpetrators with crime scenes. It wasnt employed in the United States until 1987. DNA profiling has changed forensic science. DNA technology has given police and the courts a means of identifying the suspects of rapes and murders. Thousands of cases have been closed and innocent suspects freed with guilty ones punished because of the power of a silent biological witness at the crime scene. Today, the Federal Bureau of Investigation performs the bulk of the forensic DNA typing for local and state law enforcement agencies. In criminal investigations, DNA from samples of hair, bodily fluids or skin at a crime scene is compared with those obtained from suspected suspects. (http://faculty.ncwc.edu) ‘DNA fingerprinting, or DNA typing (profiling) as it is now known, was first described in 1985 by an English genet icist named Alec Jeffreys. Dr Jeffreys found that certain regions of DNA contained DNA sequences that were repeated over and over again next to each other. Since that time, human identity testing using DNA typing methods has been widespread. The past 25 years have seen tremendous growth in the use of DNA evidence in crime scene investigations as well as paternity testing. Today over 100 public forensic laboratories and several dozen private paternity testing laboratories conduct hundreds of thousands of DNA tests annually in the United States. In addition, most countries in Europe and Asia have forensic DNA programs. The number of laboratories around the world conducting DNA testing will continue to grow as the technique gains in popularity within the law enforcement com munity. (John Butler, 2005) How DNA Typing Is Done Only one-tenth of a single percent of DNA (about 3 million bases) differs from one person to the next. (Internet Source www.ornl.org, 2009) Scientists can use these variable regions to generate a DNA profile of an individual, using samples from blood, bone, hair, and other body tissues and products. In criminal cases, this generally involves obtaining samples from crime-scene evidence and a suspect, extracting the DNA, and analyzing it for the presence of a set of specific DNA regions (markers). Scientists find the markers in a DNA sample by designing small pieces of DNA (probes) that will each seek out and bind to a complementary DNA sequence in the sample. A series of probes bound to a DNA sample creates a distinctive pattern for an individual. Forensic scientists compare these DNA profiles to determine whether the suspects sample matches the evidence sample. A marker by itself usually is not unique to an individual; if, however, two DNA samples are alike at four or five regions, o dds are great that the samples are from the same person. If the sample profiles dont match, the person did not contribute the DNA at the crime scene. If the patterns match, the suspect may have contributed the evidence sample. While there is a chance that someone else has the same DNA profile for a particular probe set, the odds are exceedingly slim. Many judges consider this a matter for a jury to take into consideration along with other evidence in the case. RFLPs can be used determine the disease status of an individual. RFLPs can be used to measure recombination rates which can lead to a genetic map with the distance between RFLP loci measured in centiMorgans. Next, the PCR product is purified and subjected to restriction enzyme digestion with enzymes that have 4 base pair recognition sites. This step generates fluorescently-labeled terminal restriction fragments. The digested products are then se parated and detected on an appropriate electrophoresis platform. For a given sample the terminal fragments will contain a fluorescent label at the 5 end and will therefore be detected. The output will be a series of peaks (fragments) of various sizes and heights that represents the profile of that sample. (Osborn, A. M., Moore, R.B. and Timmis, K.N., 2000) Polymerase chain reaction PCR is used to make millions of exact copies of DNA from a biological sample. DNA amplification with PCR allows DNA analysis on biological samples as small as a few skin cells. A polymerase is a naturally occurring enzyme, a biological macromolecule that catalyzes the formation and repair of DNA (and RNA). The technique was made possible by the discovery of Taq polymerase, the DNA polymerase that is used by the bacterium Thermus auquaticus that was discovered in hot springs. This DNA polymerase is stable at the high temperatures need to perform the amplification, whereas other DNA polymerases become denatured. Since this technique involves amplification of DNA, the most obvious application of the method is in the detection of minuscule amounts of specific DNAs. This is important in the detection of low level bacterial infections or rapid changes in transcription at the single cell level, as well as the detection of a specific individuals DNA in forensic sci ence. It can also be used in DNA sequencing, screening for genetic disorders, site specific mutation of DNA, or cloning or subcloning of cDNAs. Variability in STR regions can be used to distinguish one DNA profile from another. The Federal Bureau of Investigation (FBI) uses a standard set of 13 specific STR regions for CODIS. CODIS is a software program that operates local, state, and national databases of DNA profiles from convicted offenders, unsolved crime scene evidence, and missing persons. The odds that two individuals will have the same 13-loci DNA profile is about one in a billion. (Internet Source www.ornl.org, 2009) The Federal Bureau of Investigation (FBI) has chosen 13 specific STR loci to serve as the standard for CODIS. The purpose of establishing a core set of STR loci is to ensure that all forensic laboratories can establish uniform DNA databases and, more importantly, share valuable forensic information. If the forensic or convicted offender CODIS index is to be used in the investigative stages of unsolved cas es, DNA profiles must be generated by using STR technology and the specific 13 core STR loci selected by the FBI. (Internet Source www.dna.gov, 2009) Mitochondrial DNA analysis mtDNA can be used to examine the DNA from samples that cannot be analyzed by RFLP or STR. Nuclear DNA must be extracted from samples for use in RFLP, PCR, and STR; however, mtDNA analysis uses DNA extracted from another cellular organelle called a mitochondrion. (Internet Source www.fbi.gov, 2009) While older biological samples that lack nucleated cellular material, such as hair, bones, and teeth, cannot be analyzed with STR and RFLP, they can be analyzed with mtDNA. In the investigation of cases that have gone unsolved for many years, mtDNA is extremely valuable. (Internet Source www.dna.com, 2009) All mothers have the same mitochondrial DNA as their offspring. This is because the mitochondria of each new embryo come from the mothers egg cell. The fathers sperm contributes only nuclear DNA. Comparing the mtDNA profile of unidentified remains with the profile of a potential maternal relative can be an important technique in missing-person investigations. ( Melton, T. et. al., 2001) Y-Chromosome Analysis The Y chromosome is passed directly from father to son, so analysis of genetic markers on the Y chromosome is especially useful for tracing relationships among males or for analyzing biological evidence involving multiple male contributors. Y chromosome analysis is a useful technique for analyzing DNA that can be likened in one sense to studying male surnames. Think about the way that male surnames are passed down from one generation to another and continue on through sons. This mechanism is a simplistic representation of Y chromosomes. A son inherits a Y chromosome from his biological father and he also inherits an X chromosome from his biological mother. Conversely, a female would inherit an X chromosome from her biological mother and an X chromosome from her biological father. This type of DNA analysis has important ramifications for scientists wishing to investigate the familial ties between male members. DNA technology affords the forensic scientist the ability to eliminate individuals who have been falsely associated with a biological sample and to reduce the number of potential contributors to a few (if not one) individuals. Inculpations are strong evidence regarding the source of the biological sample. Today, some wrongly convicted people have been exonerated because of DNA evidence. Moreover, in casework, individuals are excluded routinely. Since the inception of forensic DNA profiling, there has been a debate in the legal setting regarding admissibility on the methods and the practices of computing DNA profile frequencies. While the scientific basis of DNA typing were sound, both the methodology and the statistical interpretations were aggressively challenged in court. The methods challenge focused on reliability and validity testing. The statistics debate focused on the reliability of the ass umption of independence for applying the product rule to derive estimates of DNA profile frequencies.
